PPES Service Facility for Global Kinase Activity Profiling
The University of Antwerp PPES-kinome service facility uses the phospho-peptide PamGene platform for simultaneous analysis of multiple protein kinases activities in cells, tissues and body fluids lysates by profiling peptide phosphorylation changes, using a pharmacology-on-chip approach. It allows researchers active in the areas of fundamental science, translational research, and diagnostics to gain insights in discriminative drug response, kinase inhibitor activity and discovery of novel biomarkers, targets and pathways.
Situation before
Protein phosphorylation is a reversible post-translational modification playing a crucial role in controlling the functional state of proteins in health and disease. Profiling protein kinase activity has become a critical component in molecular clinical biology research and constitutes a promising tool for drug discovery. However, the existing strategies can monitor only single kinases or imply complex procedures and infrastructure difficulting their assessment.
Technology
The PPES platform performs parallel measurements of kinase activities by recording phosphorylation changes in real time, revealing a more realistic view of the cellular signaling and molecular mechanisms operating in disease and drugs mode of action.

About the researchers/research group
Research in PPES's lab combines advanced molecular, biochemical pharmacology, Kinome, (chemo)proteomic, transcriptomic and epigenomic systems biology approaches to identify biomarkers and druggable receptor-signaling targets for the control and monitoring of therapeutic responses in cancer, infectious, neurologic, metabolic, and chronic inflammaging diseases. The lab also investigates the therapeutic or toxicological properties of medicinal (Phyto)therapeutics, hormones, metabolites and non-thermal plasma in different in vitro and in vivo disease models.
